I think in the episodes he is always wearing it. But this seems to be either a deleted scene or one that was rehearsed but not filmed. So I wonder if there was a purpose to him not wearing the ring.

I think it was one deduction too many? At the beginning of HLV I get the impression that he is neither really happy nor fulfilled in his marriage, e.g. the dream of Sherlock and the ensuing quarrel. The aggression bubbling up in the scene in the crack den. This is not a happily married man enjoying life in Suburbia. 

He cycles to the workplace he shares with Mary while she is going by car. He has his shirts readily folded like something you would pack into a suitcase. And Billy Wiggins sees it all. And we can assume that Sherlock sees it, too.

This is what we get in the scene. It is a sort of game between Sherlock and Billy who both deduce John and are throwing the balls to each other. Not sure if it is just meant as a joke. Of course it makes sense that he folds his shirts and puts them in a backpack because he is cycling to work to stay fit. This is the superficial interpretation. But we also get the hint about "the addict in need of a fix", i.e. John, and the fix being danger/adventure/Sherlock. So there is a deeper meaning to the whole scene. 
Based on what we get before to me it signals that John does not get everything he needs from his new life. As simple as that.

He has his shirts folded, not on hangers, so that he can pack them anytime into a backpack or suitcase. That is all I can say.
