The one quote that randomly pops into my head and gets me giggling is "Sherlock Holmes, if you've been YouTubing!"

There's so much revealed about both Mrs. H and Sherlock in that quote, I don't know where to start.

Also love the bit in HLV when John finds Sherlock in the drugs den and he bursts through the door before bellowing "WELL I'M NOT NOW!" and poor Mary is in the car all confused haha!

Also very funny: Sherlock asks Lestrade to send over his least irritating officer - and it turns out that Lestrade's least irritating officer is Lestrade himself.

I've just watched TSOT and thought it was so funny when inebriated Sherlock looks at things and sees what we see (like "cardigan" and "sleep"), instead of his usual brilliant deductions. He gets a glimpse of what it's like inside our tiny brains.
